The invention relates to a nano-barrier additive having a structure as shown in Formula I, wherein POSS has a structure as shown in Formula II, and R has a structure as shown in Formula III. The invention also relates to a nano-barrier polylactic acid masterbatch comprising the following components by mass percentage: 73%–80.9% polylactic acid, 13%–17% nano-barrier additive, 0.1%–1% antioxidant, 0.5%–2% dispersant, and 3%–10% compatibilizer. The invention further relates to a preparation method of a polylactic acid packaging film: first, mixing and extruding polylactic acid, nano-barrier additive, antioxidant, dispersant and compatibilizer for granulation to obtain a nano-barrier polylactic acid masterbatch; then extruding and blow-molding the nano-barrier polylactic acid masterbatch with polylactic acid, followed by heat setting. The nano-barrier additive of the invention can effectively block water vapor, and the polylactic acid masterbatch using the nano-barrier additive exhibits excellent mechanical properties.
